The two were found unconscious below the bridge, which is close to the Star public house near the A5. They had fallen in a stream which runs into the River Dee.
Police said the incident happened shortly after midnight yesterday. Inspector Tony Latham said the emergency services were called to a report of a male and a female having fallen over the side wall of a bridge on Queens Street, Llangollen.
He said that both were taken to hospital and were receiving treatment.
"One has sustained serious injuries," he said.
A spokesman for Welsh Ambulance said two ambulances went to the scene.
He said: "A 21-year-old female was unconscious and in a serious condition.
"The male had a fractured arm. We took them both to the Wrexham Maelor Hospital."
The police are not treating the matter as suspicious, but Inspector Latham said: "We are appealing for anyone who may have witnessed the incident to telephone the Force Control room 0845 607 1002 (English line) quoting incident reference L085020."
