The 114-miles route will end at Caerphilly Castle, making the whole race a 'castle to castle' event. A total 96 riders from 16 teams are taking part, including six British teams, as well as Irish, American, Italian, German, French, Dutch and Belgian teams.
Julian Winn, manager of the Endura Racing team and gold medallist at the 2002 Commonwealth Games, said that Powis Castle would make a great backdrop to start the race.
"All along the route there is some stunning scenery and Powis Castle should allow for some great aerial views," he said.
"Cycling in general is on a roll in Wales at the moment.
"A race as big as this is a good way to get people interested in cycling."
The tour got under way in Scotland on Sunday, and stage five will see competitors racing between Exeter and Exmouth in Devon.
