The competition, for teams in the Scottish Championship and Scottish Leagues One and Two, is being extended and revamped for next season.
TNS and fellow Welsh Premier side Bala Town will join two Northern Ireland clubs, Crusaders and Linfield, plus four teams each from the Highland and Lowland Leagues.
In addition, Scottish top-flight clubs will each field an under-20 side.
TNS chief operating officer Ian Williams said: "We are thrilled to be involved in the cup and look forward to competitive matches."
