The 22-year-old from Hawkstone Park was beaten by one hole by Austrian teenager Matthias Schwab in the first round of the matchplay stages at Royal Troon
It was a bitter pill for Chesters to swallow as he looked destined to reach the last 32 of one of his biggest tournaments of the year as he was one up at the 17th tee.
But successive bogeys at the last two holes proved costly for the Shropshire ace, who saw his hopes of emulating last year's run to the last 16 dashed.
"I played really well until the last two holes," said Chesters. "It's really disappointing because I was confident I could go further than last year."
Chesters had made a flying start to the week by carding the joint-best score of the 288-strong field in Monday's opening day of strokeplay qualifying, a four-under-par 67 at Glasgow Gailes.
He followed that up with a 75 at Troon on Tuesday to comfortably qualify for the matchplay stages on even par in a share of 15th spot.
Chesters now heads to Meir on Monday for the first stage of Open qualifying before competing in the Brabazon Trophy – the English Strokeplay Championship.
Ludlow's Oliver Farr continued his impressive form in the latest EuroPro Tour event.
He carded a four-under-par 68 at the WSL Open at Collingtree Park to lie in a share of 10th spot with two rounds remaining.
